Дослідження рекомбінації між плечами 1RS від жита Petkus та Insave у складі пшенично-житніх транслокацій 1BL.1RS і 1AL.1RS з використанням запасних білків як генетичних маркерів<

Создана популяция рекомбинантно-инбредных линий F6 от скрещивания линий озимой мягкой пшеницы с пшенично-ржаными транслокациями 1BL.1RS (типа Кавказ) и 1АL.1RS (типа Amigo) Б-16 × 7086 AR. С использованием запасных белков, контролируемых локусами Gli-R1, Gli-A1, Gli-B1, как генетических маркеров идентифицированы линии с рекомбинантными 1RS (12 %), и определена частота рекомбинации между плечами 1RS в составе разных транслокаций на уровне 7 %. Показано, что 1RS от Amigo имеет ген, кодирующий секалин ‘a’, который можно идентифицировать на SDS-электрофореграмме под y-субъединицей, кодированной локусом Glu-D1. Cекалин ‘a’ также обнаружен у сорта MV Táltos, у которого идентифицирована транслокация 1BL.1RS с аллелями секалиновых локусов типа Amigo. Показано, что ген, кодирующий секалин ‘a’ от Amigo, и ген Sec-Nx от ржи Воронежская СГИ являются аллельными. Частота рекомбинации между локусом Gli-R1 и Sec-N зависит от природы проанализированного материала и составляет около 10 % (расстояние – 10 сМ) в скрещивании MV Táltos × CWX (линии с разными транслокациями 1BL.1RS). Такое расстояние позволяет предположить, что важные гены устойчивости против возбудителей болезней и вредителей, в частности устойчивости против возбудителя стеблевой ржавчины, находятся на участке между этими локусами. Поэтому секалиновые локусы являются удобными для первичного скрининга материала с рекомбинантными плечами 1RS с новыми комбинациями генов устойчивости про-тив возбудителей болезней и вредителей.

Створено популяцію рекомбінантно-інбредних ліній F6 від схрещення ліній озимої м’якої пшениці з пшенично-житніми транслокаціями 1BL.1RS (типу Кавказ) і 1АL.1RS (типу Amigo) Б-16 × 7086 AR. З використанням запасних білків, контрольованих локусами Gli-R1, Gli-A1, Gli-B1, як генетичних маркерів, ідентифіковано лінії з рекомбінантними 1RS (12 %), та визначено частоту рекомбінації між плечами 1RS у складі різних транслокацій на рівні 7 %. Показано, що 1RS від Amigo має ген, що кодує секалін ‘a’, який можна ідентифікувати на SDS-електрофореграмі під y-субодиницею, кодованою локусом Glu-D1. Cекалін ‘a’ також виявлено у сорту MV Táltos, у якого ідентифіковано транслокацію 1BL.1RS з алелями секалінових локусів типу Amigo. Показано, що ген, що кодує секалін ‘a’ від Amigo, та ген Sec-Nx від жита Воронезьке СГІ є алельними. Частота рекомбінації між локусом Gli-R1 та Sec-N залежить від природи проаналізованого мате-ріалу і становить біля 10 % (відстань – 10 сМ) у схрещенні MV Táltos × CWX (лінії з різними транс-локаціями 1BL.1RS). Така відстань дозволяє припустити, що важливі гени стійкості проти збудників хвороб і шкідників, зокрема стійкості проти збудника стеблової іржі, знаходяться на ділянці між цими локусами. Тому секалінові локуси є зручними для первинного скринінгу матеріалу з рекомбінантними плечами 1RS з новими комбінаціями генів стійкості проти хвороб і шкідників.
