Histological peculiarities of the morphogenesis in Triticum aestivum L. callus cultures under the influence of antibiotic ceftriaxone

Показано, что для эффективного получения регенерантов у мягкой пшеницы целесообразно использовать 18-суточный каллус, образованный из апикальных меристем трехдневных асептических проростков. В нем наблюдаются пролиферативные центры с развитыми элементами проводящей системы, способствующие появлению меристематических очагов, из которых могут формироваться регенеранты. Показано, что регенерация из каллуса идет путем прямого органогенеза, а не соматического эмбриогенеза. Под действием антибиотика цефтриаксона, добавленного в регенерационную среду в концентрации 400 мг/л, у двух разных генотипов мягкой пшеницы наблюдается, во-первых, изменение морфологии каллуса апикального происхождения, во-вторых, ускорение регенерационных процессов, в-третьих, образование корней у побегов, что позволяет избежать дополнительного эта-па укоренения растений.

SUMMARY. It was shown that for efficient production of regenerants in bread wheat, 18-day old calli, obtained from apical meristems of 3-day-old aseptic seedlings should be used. These calli show proliferative centers, with developed vascular system elements, that support the development of meristematic centers which later produce regenerants. It was shown that regeneration from the calli occurs by direct organogenesis but not by somatic embryogenesis. Under the influence of the antibiotic ceftriaxone, added to the regeneration medium in concentration of 400 mg/l, two different bread wheat genotypes showed, firstly, changes in apical origin callus morphology; secondly, an intensification of the regeneration processes; thirdly, roots development in the regenerants, thereby it allows to avoid the additional step of plants rooting.
