The RIB7 gene encoding riboflavin synthase of Pichia guilliermondii has been cloned on the vector pBR322 as the 3.7 kbp HindIII and 12.0 kbp EcoRI fragments of the chromosomal DNA. The structural gene was localized in the central region of the cloned EcoRI fragment. The constructed plasmids complemented riboflavin deficiency in a yeast mutant strain defective in riboflavin synthase, but complete recovery of the wild type phenotype was only observed when linear DNA fragments carrying the RIB7 gene were used for transformation. The peculiarities of recombination of hybrid plasmids in yeast P. guilliermondii are discussed.
