IRAP-аналіз трансгенних рослин пшениці з дволанцюговим РНК-супресором гена проліндегідрогенази

РЕЗЮМЕ. Методом IRAP анализа проанализирован уровень полиморфизма участков ДНК, фланкированных инвертированными LTR повторами ретротранспозонов, у генетически модифицированных растений пшеницы, содержащих двухцепочечный РНК-супрессор гена пролиндегидрогеназы, полученных методом Agrobacterium-опосредованной трансформации в культуре in vitro. При использовании всех высокоэффективных праймеров к ретротранспозонам Sukkula, Sabrina, Wham, Nikita и Wilma1 у трансгенных растений полиморфизма ДНК обнаружено не было. В условиях проведенного эксперимента нами не зарегистрировано исчезновение ампликонов в ДНК профилях ПЦР, что может свидетельствовать об отсутствии перестроек в сайтах связывания с праймером и в исследуемых локусах, фланкированных LTR ретротранспозонов. В спектрах продуктов амплификации ДНК не отмечено появления новых ампликонов, что свидетельствует об отсутствии активации транспозиционной активности мобильных генетических элементов у изученых трансгенных растений с двухцепочечным РНК-супресором гена пролиндегидрогеназы. Для расширения спектра амп-ликонов в продуктах ПЦР исследуемых образцов была испытана методика сочетания IRAP-праймеров к различным ретротранспозонам в одной реакции. Экспериментальным путем были подобраны пары IRAP-праймеров, однако и при использование этого способа не было обнаружено исчезновения или появления новых полиморфных фрагментов. Отсутствие полиморфизма ДНК у трансгенных растений с двухцепочечным РНК-супресором гена пролиндегидрогеназы возможно может быть связано с явлением РНК-интерференции, которая подавляет активность ретротранспозонов.

Методом IRAP аналізу проаналізовано рівень поліморфізму ділянок ДНК, фланкованих інвертованими LTR повторами ретротранспозонів, у генетично-модифікованих рослин пшениці, що містять дволанцюговий РНК-супресор гена проліндегідрогенази, отриманих шляхом Agrobacterium-опосередкованої трансформації в культурі in vitro. За використання високоефективних праймерів до ретротранспозонів Sukkula, Sabrina, Wham, Nikita та Wilma1 у трансгенних рослин поліморфізму ДНК виявлено не було. За умов проведеного експерименту нами не зареєстровано зникнення ампліконів у ДНК профілях ПЛР, що може свідчити про відсутність перебудов у сайтах зв’язування з праймером та в досліджуваних локусах. У спектрах продуктів ампліфікації ДНК не відмічено появи нових ампліконів, що свідчить про відсутність активації транспозиційної активності МГЕ у досліджених трансгенних рослин з дволанцюговим РНК-супресором гена проліндегідрогенази. Для розширення спектру ампліконів у продуктах ПЛР досліджуваних зразків була випробовувана методика поєднання IRAP-праймерів до різних ретротранспозонів в одній реакції. Експериментальним шляхом були підібрані пари IRAP-праймерів, проте і за використання цього способу не було виявлено зникнення або появи нових поліморфних фрагментів. Відсутність поліморфізму ДНК у трансгенних рослин з дволанцюговим РНК-супресором гена проліндегідрогенази, можливо, може бути пов’язана з явищем РНК-інтерференції, яка пригнічує активність ретротранспозонів.
