The expression of glypican­3 in colorectal cancer

РЕЗЮМЕ. Гліпікан-3 (GPC3), гепарансульфат-протеоглікан, – це маркер появи пухлини. Його підвищену експресію було встановлено для декількох видів раку, зокрема, гепатоцелюлярної карциноми (HCC), меланоми, тощо. У цьому дослідженні ПЦР-аналіз у реальному часі було використано для вивчення експресії мРНК GPC3 у 61 зразку тканин пацієнтів з колоректальним раком (CRC) та 61 зразку нормальних суміжних тканин. За допомогою імуногістохімії вивчали експресію білка GPC3 у випадках значного підвищення експресії рівня мРНК GPC3 у тканинах пухлин порівняно з нормальними тканинами. Суттєве підвищення рівня експресії гену GPC3 було виявлено у 49,2 % пухлин колоректального раку порівняно з нормальними суміжними тканинами. Було встановлено, що експресія мРНК GPC3 суттєво корелює з патологічною диференціацією та віком пацієнтів. Варто відмітити, що значне зниження експресії мРНК GPC3 було виявлено у тканинах пухлин 16 пацієнтів, які пройшли неоад’ювантну хіміотерапію, порівняно з нормальними суміжними тканинами. Неочікувано, білок GPC3 не було виявлено за допомогою імуногістохімії у 30 тканинах, що продемонстрували підвищення експресії мРНК GPC3. Наші результати вказують на те, що експресія GPC3 можлива у тканинах колоректального раку на рівні мРНК, тоді як на рівні білку цієї експресії виявлено не було.
