Получение трансгенных растений картофеля, экспрессирующих ген лактоферрина человека, и анализ их устойчивости к фитопатогенам

С помощью Agrobacterium-опосредованной трансформации осуществлен перенос гена лактоферрина человека (hLf) в геном ряда сортов картофеля (Solanum tuberosum) украинской селекции (Вернисаж, Левада, Свитанок Киевский и Зарево). Для этого использовали плазмидный вектор pBIN35LF, несущий ген hLf под контролем 35S промотора вируса мозаики цветной капусты и терминатора октопинсинтазы, а также селективный маркерный ген неомицинфосфотрансферазы ІІ (nptII), обеспечивающий устойчивость к канамицину. В результате селекции были отобраны 44 линии сорта Вернисаж, 26 линий сорта Левада, 25 линий сорта Свитанок Киевский и 16 линий сорта Зарево, устойчивых к 100 мг/л канамицину. Интеграция целевого гена в геном картофеля была подтверждена с помощью полимеразной цепной реакции с использованием специфических праймеров к гену hLf. Идентификацию рекомбинатного лактоферрина в тканях трансгенных линий проводили с помощью Вестерн блоттинга с использованием специфических моноклональных антител к лактоферрину. Отобранные трансгенные линии картофеля были протестированы на устойчивость к бактериальным и грибному фитопатогенам. С помощью теста диффузии в агар установлено, что сок трансгенных линий картофеля обладает антибактериальным эффектом по отношению к таким фитопатогенным бактериям, как Ralstonia solanacearum (возбудитель бактериальной гнили картофеля) и Clavibacter michiganensis subsp. sepedonicus (возбудитель кольцевой гнили картофеля). Устойчивость трансгенных растений картофеля к фитофторозу изучали путем заражения растений in vitro изолятом Phytophthora infestans. В результате была установлена повышенная устойчивость полученных трансгенных линий картофеля к P. infestans по сравнению с контролем. Таким образом, полученные данные показывают, что перенос гена hLf гена в геном картофеля повышает его устойчивость к бактериальным и грибным патогенам.

РЕЗЮМЕ. За використання Agrobacterium-опосередкованої  трансформації було здійснено перенесення  гена лактоферина людини в геном ряда сортів картоплі (Solanum tuberosum) української селекції (Вернісаж, Левада, Світанок Київський та Зарево). Для цього використовували плазмідний вектор pBIN35LF, який ніс ген лактоферина людини hLf під контролем 35S промотора віруса мозаїки цвітної капусти та термінатора октопінсинтази, а також селективний маркерний ген неоміцинфосфотрансферази ІІ (nptII), що забезпечує стійкість до канаміцина. В результаті селекції були відібрані 44 лінії сорта Вернісаж, 26 ліній сорта Левада, 25 ліній сорта Світанок Київський та 16 ліній сорта Зарево, стійких до 100 мг/л канаміцина. Інтеграція цільового гена в геном картоплі була підтверджена за допомогою метода полімеразної ланцюгової реакції за використання специфічних праймерів до гена hLf. Ідентифікацію рекомбінатного лактоферина в тканинах трансгенних ліній проводили за допомогою Вестерн блоттинга за використання специфічних моноклональних антитіл до лактоферину. Відібрані трансгенні лінії картоплі були протестовані на стійкість до бактеріальних та грибних фітопатогенів. За допомогою теста дифузії в агар було встановлено, що сік трансгенних ліній картоплі володіє антибактеріальним ефектом по відношенню до таких фітопатогенних бактерій, як  Ralstonia solanacearum (збудник бактеріальної гнилі картоплі) та Clavibacter michiganensis subsp. sepedonicus (збудник кільцевої гнилі картоплі). Стійкість трансгенних ліній картоплі до фітофтороза досліджували шляхом зараження рослин in vitro ізолятом Phytophthora infestans. В результаті була встановлена підвищена стійкість отриманих трансгенних ліній картоплі до P. infestans у порівнянні з контролем. Таким чином, отримані дані вказують на те, що перенос гена hLf в геном картоплі підвищує її стійкість до бактеріальних та грибних патогенів.
