Enhanced in vitro regeneration in sugarcane (Saccharum officinarum L.) by use of alternate high-low picloram doses and thidiazuron supplementation

РЕЗЮМЕ. Цукрова тростина – це важлива рослина для внутрішнього використання та промислового виробництва в країні. Було докладено значних зусиль для покращення її продуктивності за допомогою традиційних та біотехнологічних підходів. Генетичні маніпуляції з цукровою тростиною залежать від ефективності відтворюваного режиму in vitro регенерації. У цьому дослідженні оцінювали роль положення експланта, індукованого пересихання, рівнів пікло-раму (PIC) у калюсогенезі, а також важливість додавання тідіазурону (TDZ) впродовж фази регенерації. Оптимальне поєднання вищевказаних факторів дозволило визначити покращену систему in vitro регенерації для двох відбірних сортів цукрової тростини. Ембріогенез сорту HSF-240 стимулювали за допомогою MS середовища з додаванням 12,42 мкМ PIC, у той час як сорт CPF-237 продемонстрував соматичний ембріогенез, коли додавання PIC поєднували з індукованим пересиханням (за допомогою 12 г л–1 агару). Було зафіксовано зниження частоти ембріогенезу від основи до верхівки. Експланти, вирощені змінно на середовищі з високим (12,42 мкМ) і низьким (4,14 мкМ) вмістом PIC, виробили найбільшу кількість нодулярних калюсів, які згодом показали максимальний регенераційний потенціал. Оптимальне формування пагонів спостерігали при 9,08 мкМ TDZ, середні значення були зафіксовані при 2,27 мкМ 2,4-дихлорфеноксиоцтової кислоти з додаванням 4,43 мкМ бензиламінопурину. Однак, пагони, сформовані з першим складом середовища, продемонстрували ламкість порівняно з пагонами, регенерованими на другому середовищі. Здорові корені формувалися за допомогою 16,11 мкМ нафталіноцтової кислоти у присутності 0,5 % активованого вугілля. Вміст малонового диальдегіду, активність каталази і пероксидази in vitro та у рослин цукрової тростини, вирощених у польових умовах, були аналогічними, що вказує на те, що in vitro регенеровані рослини були рівноцінно здатними до подальшого вирощування у природних умовах. Заявлений протокол може бути корисним для розробки стратегій щодо повноцінного режиму генетичної інженерії цукрової тростини.
