Inflammatory and anti­inflammatory cytokine activity in the cartilage cells of genetically modified mice

РЕЗЮМЕ. Було досліджено рівень активності різних генів у моношарових культурах хондропрогеніторних клітин і хондроцитів, виділених з суглобових хрящів генетично модифікованих мишей. Дослідження моношарових культур хондроцитів і хондропрогеніторних клітин було проведене за допомогою кількісної ПЛР. Метою дослідження було визначення активності цільових генів: col 2a1, Sox 9, Il 1a, Il 1b, CCL 2, CCL 3, CCL 4, CCL 5, MMP 3, MMP 13 і аггрекану (Aggr). Було виявлено, що активність прозапальних цитокінів (Il1b, Il6, Il 8) є вищою у хондропрогеніторних клітинах. Активність метало­протеїназ (MMP 3, MMP 13) є причиною деградації матриці. Синтез колагену типу 2 і аггрекану у хондропрогеніторних клітинах є інтенсивнішим, аніж у хондроцитах, подібне спостерігається у всіх штамах тварин, що вивчалися. Було відмічено високий рівень активності колагену типу 2 і аггрекану у хондропрогеніторних клітинах і хондроцитах штаму 6, виділеного з хрящової тканини, водночас спостерігався низький рівень активності прозапальних цитокінів і металопротеїназ, що вказує на значну здатність цього штаму мишей до репарації пошкодженої хрящової тканини.
