Дослідження субпопуляції стовбурових пухлинних клітин на моделях раку молочної залози

Гетерогенна природа пухлинної популяції, наявність стовбурових пухлинних клітин, є однією з причин стійкості злоякісних новоутворень до протипухлинної терапії, виникнення рецидивів та метастазів а також складності лікування даної патології. Метою роботи було збагатити багатоклітинні пухлинні сфероїди клітин аденокарциноми молочної залози лінії MCF-7 стовбуровими пухлинними клітинами та дослідити отриману субпопуляцію стовбурових пухлинних клітин за допомогою біохімічних, імунологічних та цитологічних методів. За результатами дослідження було з’ясовано, що в умовах збіднілого поживного середовища, при додаванні певних факторів росту, відсоток стовбурових пухлинних клітин в популяції багатоклітинних пухлинних сфероїдів суттєво збільшується. Так, відсоток клітин, які експресують CD133 збільшується від 12,47 до 82,08 %, Nestin від 31,3 до 82,58 %. Підвищується також кількість клітин, що експресують маркери стовбурових пухлинних клітин: CD44, CD133, bmi1, згідно даних імуноцитохімічного забарвлення. Активність альдегід дегідрогенази в клітинах лінії MCF-7 за умов моношарового росту складала 0,07 моль/мг білка за хв та збільшувалася до 1,58 моль/мг білка за хв у збагачених на стовбурові пухлинні клітини багатоклітинних пухлинних сфероїдах. Активність глюкозо-6-фосфат дегідрогенази в пухлинних клітинах за умов моношарового росту становила 934,6 ± 148,3 × 10–6 моль/мг білка за хв при збагаченні багатоклітинних пухлинних сфероїдів на стовбурові пухлинні клітини активність глюкозо-6-фосфат дегідрогенази зростала у більш ніж у 1,5 рази. Активність лактатдегідрогенази в клітинах MCF-7 за умов моношарового росту становила 65,12 ± 1,28 мкмоль/мг білка за хв, а за умов росту в збагачених стовбуровими клітинами багатоклітинних пухлинних сфероїдах, зменшувалася у 5,5 разів. Таким чином, за результатами аналізу отриманих даних, ми можемо при-пустити, що за умов збагачення пухлинної популяції на стовбурові пухлинні клітини змінюється рецепторний та енергетичний профіль клітин MCF-7, наближаючи збагачені стовбуровими пухлиними клітинами сфероїди до характеристик метастазуючого мікровузла, а пухлинні клітини до стовбурових пухлинних клітин.
