Early evidence of global dna methylation and hydroxymethylation changes in rat kidneys consequent to hyperoxaluria-induced renal calcium oxalate stones

РЕЗЮМЕ. Надлишковий вміст кальцію в сечі внаслідок гіпероксалурії може призводити до утворення ренальних кальцій-оксалатних каменів (CaOx), тобто нефролітіазу. Нещодавні генетичні дослідження виявили вірогідність епігенетичних змін під час нефролітіазу. Наша мета полягала у вивченні впливу утворення ренальних CaOx каменів на загальні рівні 5-метилцитозину (5 mC) і 5-гідрометилцитозину (5hmC) у нирках щурів, уражених гіпероксалурією. У цьому дослідженні дванадцять самців щурів лінії Вістар поділили порівну на дві групи (контрольну і експериментальну). Тварини в експериментальній групі отримували 2,5 % (м/об) оксалату натрію в 5 мл стерильної води перорально щодня протягом чотирьох тижнів для індукції гіпероксалурії. Наприкінці чотирьох тижнів провели оцінку параметрів сечі, біохімічних показників сироватки та ренальних гістопатологічних змін. Було проаналізовано загальні рівні 5 mC та 5 hmC за допомогою імуноферментного аналізу (ELISA) та встановлено ренальну мРНК експресію генів DNMTs та TET. Параметри сечі і біохімічні показники сироватки продемонстрували, що після перорального отримання 2,5 % (м/об) оксалату натрія протягом чотирьох тижнів у щурів розвинулась гіпероксалурія (p < 0,05). Гістопатологічна оцінка нирок щурів, вражених гіпероксалурією, показала відкладення кристалів CaOx та виражене враження канальців нирок. Результати ELISA продемонстрували значно вищі рівні 5hmC (p < 0,0001) у нирках щурів з ренальними CaOx каменями у той час, як рівні 5mC були менш значними (p < 0,05) порівняно з щурами у контролі. Було виявлено, що рівень експресії мРНК TET 2, регулятора процесу деметилювання ДНК, був значно підвищений (p < 0,01) у щурів з CaOx каменями в нирках. Це попереднє дослідження продемонструвало ранні докази епігенетичних модифікацій під впливом індукованих гіпероксалурією ренальних CaOx каменів, що може сприяти транскрипційній регуляції під час CaOx нефролітіазу.
