Структурно-биологическая характеристика изотипов СК1-подобных протеинкиназ, участвующих в фосфорилировании микротрубочек растений

Идентифицировано 18 изотипов CK1-подобных протеинкиназ у A. thaliana. Сравнение каталитических доменов протеинкиназ CK1 крысы (α, β, γ1-3, δ и ε) и 18-ти гомологов из A. thaliana подтвердило значительное структурное сходство 13-ти протеинкиназ: CKL1 (CK1δ), CKL2, CKL3, CKL4, CKL5, CKL6, CKL7, CKL8, CKL9, CKL10, CKL11, CKL12 и CKL13. Было обнаружено, что CK1-специфический ингибитор D4476 взаимодействует с KC1D крысы и отобранными растительными гомологами аналогичным АТФ-кон-курентным способом. Взаимодействие с лигандом было подтверждено значениями оценочных функций докинга, результатами молекулярной динамики и результатами хемогенного анализа. Известно, что взаимодействие протеинкиназ CK1 с субстратными белками в значительной степени зависит от наличия специфических мотивов, расположенных в C-концевой области. Соответствующие мотивы взаимодействия с EB1 были выявлены в С-концевых областях СKL1 и CKL2. Также была подтверждена роль С-коневого фрагмента CKL6 во взаимодействии с β-тубулином.

РЕЗЮМЕ. Ідентифіковано 18 ізотипів CK1-подібних протеїн-кіназ у A. thaliana. Порівняння каталітичних доменів протеїнкіназ CK1 пацюка (α, β, γ1-3, δ і ε) і 18-ти гомологів з A. thaliana підтвердило значну структурну подібність у випадку 13-ті протеїнкиназ: CKL1 (CK1δ), CKL2, CKL3, CKL4, CKL5, CKL6, CKL7, CKL8, CKL9, CKL10, CKL11, CKL12 і CKL13. Було встановлено, що CK1-специфічний інгібітор D4476 взаємодіє з KС1D пацюка і відібраними рослинними гомологами у аналогічної АТФ-конкурентної манери. Взаємодію з лігандом було підтверджено значеннями оціночних функцій докінгу, результатами молекулярної динаміки і результатами хемогенного аналізу. Відомо, що взаємодія протеїнкіназ CK1 з субстратними білками в значній мірі залежить від наявності специфічних мотивів, розташованих в C-кінцевий ділянці. Відповідні мотиви що відповідають за взаємодію з EB1, були знайдені в С-кінцевих ділянках СKL1 і CKL2. Також була підтверджена роль С-кінцевого фрагмента CKL6 у взаємодії з β-тубуліном.
