Філогенетичний аналіз штаму Bacillus subtilis IFBG МК-2 та продукування рибофлавіну його індукованими клонами

Метою роботи було дослідження філогенетичних взаємозв’язків нового штаму-продуценту Bacillus subtilis IFBG МК-2 з вже існуючими штамами і підвищення біосинтетичної активності цього продуценту шляхом мутагенезу. В результаті сиквенування 16S рРНК штаму IFBG МК-2 отримано фрагменти цього гена, сумарною довжиною 1305 нуклеотидів. Проведено філогенетичний аналіз і побудовано дендрограму на основі нуклеотидних послідовностей гена 16S рРНК з використанням методу зв’язування найближчих сусідів і двопараметричної моделі Кімури. Виявлено, що відсоток подібності між сумарним сиквенованим фрагментом гена 16S рРНК штаму IFBG МК-2 і депонованою у GenBank послідовністю цього гена типового штаму B. subtilis ІАМ 12118 становить 99 %. За результатами філогенетичного аналізу штам IFBG MK-2 ідентифіковано як B. subtilis. Шляхом індукованого мутагенезу з використанням УФ-опромінення і послідовною ступінчатою селекцією одержано мутантний клон B. subtilis IFBG МК-1А з підвищеною на 50 % продукцією рибофлавіну у порівнянні з вихідним штамом.
