Організація міжгенного спейсера 5S рДНК та його використання в молекулярній систематиці роду Aconitum L.

Рід Aconitum L. налічує у своєму складі велику кількість токсичних та фармацевтично-важливих рослин. Один із центрів різноманіття цього роду розташований на території Східних Карпат. У цьому регіоні зустрічається багато представників роду із нечітко визначеним таксономічним статусом, зокрема, представники комплексу A. anthora s. l. Суперечливим питанням також залишається таксономічне положення цього комплексу в межах роду, оскільки використані для філогенетичного аналізу ділянки хлоропластного та ядерного геномів виявились недостатньо мінливими. Тому актуальним залишається пошук оптимального молекулярного маркеру з високим рівнем поліморфізму в межах роду Aconitum. IGS (intergenic spacer) 5S рДНК являє собою еволюційно мінливу ділянку ядерного геному, яку раніше з успіхом використовували для реконструкції філогенезу у багатьох групах Покритонасінних. В цій роботі, використовуючи молекулярно-генетичні та біоінформатичні методи ми отримали послідовності IGS 5S рДНК для представників комплексу A. anthora та філогенетично віддалених видів роду Aconitum. Аналіз послідовностей IGS виявив, що у видів роду ця ділянка має порівняно велику довжину від 574 до 619 нп. Мінливість IGS обумовлена численними нуклеотидними замінами, тоді як короткі олігонуклеотидні індели зустрічаються лише 5′-кінці на спейсера. У IGS Aconitum знайдено чотири консервативні ділянки, дві з яких відповідають зовнішнім елементам промотора та термінатора РНК-полімерази ІІІ, тоді як функція двох інших ділянок залишається невідомою. Перша з них демонструє гомологію до кодувальної ділянки 5S рРНК, тоді як друга має високу подібність із послідовностями з геномів представників таксономічно віддалених родин одно-та дводольних рослин, що може бути наслідком горизонтального переносу генів. Філогенетичний аналіз з використанням IGS 5S рДНК підтримує трактування A. anthora s. l. в якості окремого підроду в межах роду Aconitum.
